25592 Hook Effect Prolactin

Hook Effect Prolactin
Test Code: HEPRL
Synonyms/Keywords
Hook Effect, Prolactin
Test Components
Sample will be analyzed undiluted (1:1), as well as at 1:10, 1:100 and 1:1000 dilutions. Three results will be reported as follows.
HEPRL:   Hook Effect Prolactin (Presence or Absence of Hook Effect)
PROLAC: Prolactin (Final Prolactin result reported)
PRLDIL:  Hook Effect Final Dilution
Useful For
Determine if hook effect is contributing to a Prolactin reported result.
Specimen Requirements
Fasting Required Specimen Type Preferred Container/Tube Acceptable Container/Tube Specimen Volume Specimen Minimum Volume
(allows for 1 repeat)
Pediatric Minimum Volume
(no repeat)
No Serum Serum Separator Tube (SST) Red Top Tube (RTT), Lithium-heparin Plasma Separator Tube (PST) 0.5 mL 0.4 mL 0.3 mL
Collection Processing Instructions
Samples collected in a RTT must be removed from the clot wihin 1 hour for storage or transport. Samples collected in SST or PST must be removed from the primary tube wihin 4 hours for transport.
Specimen Stability Information
Specimen Type Temperature Time
Serum/Plasma Refrigerated 48 hours
Frozen 30 days
Interference
Human Anti-Murine Antibodies (HAMA) may develop leading to test interference in patients receiving murine-based agents for chemotherapy or tumor-imaging studies.

Marshfield:  Hook Effect Prolactin
ABSENT (no hook effect)
Prolactin
Female (non-pregnant):      2.8 - 29.2 ng/mL
            (pregnant):             9.7 - 208.5 ng/mL
            (post-menopausal): 1.8 - 20.3 ng/mL

Male:     2.1-17.7 ng/mL
Interpretation

The undiluted, 1:10, and 1:100 diluted Prolactin results are compared.

ABSENT: If after correction for dilution factor all results are similar, no hook effect is suspected.  Undiluted Prolactin result is reported.

PRESENT: If the 1:10 and/or 1:100 dilutions are not similar to the undiluted sample, hook effect is likely.  Sample will continue to be diluted until similar results from one dilution to the next are observed.  Diluted Prolactin result is reported along with the final dilution performed.
